Epidemiology

Studies have shown that HPV persistent infections and multiple infections are one of the main causes of cervical cancer. Currently, the recognized effective treatments are still lack for cervical cancer caused by HPV, so early discovery and prevention of cervical infection caused by HPV is the key to preventing cervical cancer. It is of great significance to establish a simple, specific and rapid etiology diagnostic test for the clinical diagnosis and treatment of cervical cancer.

Work Flow

1. Urine sample

A: Take 1.4mL of the urine sample to be tested and centrifuge at 12000rpm for 5 minutes; discard the supernatant (it is recommended to keep 10-20μL supernatant from the bottom of the centrifuge tube), add 200μL of sample release reagent, and the subsequent extraction should be conducted according to the instructions for use of Macro & Micro-Test Sample Release Reagent (HWTS-3005-8).

B: Take 1.4mL of the urine sample to be tested and centrifuge at 12,000rpm for 5 minutes; discard the supernatant (it is recommended to keep 10-20μL of supernatant from the bottom of the centrifuge tube), and add 200μL of normal saline to resuspend, as the sample to be tested. The subsequent extraction can be conducted with Macro & Micro-Test Viral DNA/RNA Kit (HWTS-3017-50, HWTS-3017-32, HWTS-3017-48, HWTS-3017-96) (which can be used with Macro & Micro-Test Automatic Nucleic Acid Extractor (HWTS-3006C, HWTS-3006B)) by Jiangsu Macro & Micro-Test Med-Tech Co., Ltd. in strict accordance with the instructions for use. The recommended elution volume is 80μL.

C: Take 1.4mL of the urine sample to be tested and centrifuge at 12,000rpm for 5 minutes; discard the supernatant (it is recommended to keep 10-20μL of supernatant from the bottom of the centrifuge tube), and add 200μL of normal saline to resuspend, as the sample to be tested. The subsequent extraction can be conducted with QIAamp DNA Mini Kit(51304) by QIAGEN or Macro & Micro-Test Viral DNA/RNA Column (HWTS-3020-50). The extraction should be processed according to the instructions for use. The extraction sample volume is 200μL, and the recommended elution volume is 80 μL.

2. Cervical swab/vaginal swab sample

A: Take 1mL of the sample to be tested in a 1.5mL of centrifuge tube, and centrifuge at 12000rpm for 5 minutes. Discard the supernatant (it is recommended to keep 10-20μL of supernatant from the bottom of the centrifuge tube), add 100μL of the sample release reagent, and then extract according to the instructions for use of Macro & Micro-Test Sample Release Reagent (HWTS-3005-8).

B: The extraction can be conducted with Macro & Micro-Test Viral DNA/RNA Kit (HWTS-3017-50, HWTS-3017-32, HWTS-3017-48, HWTS-3017-96) (which can be used with Macro & Micro-Test Automatic Nucleic Acid Extractor (HWTS-3006C, HWTS-3006B)) by Jiangsu Macro & Micro-Test Med-Tech Co., Ltd. in strict accordance with the instructions for use. The extracted sample volume is 200μL, and the recommended elution volume is 80μL.

C: The extraction can be conducted with QIAamp DNA Mini Kit(51304) by QIAGEN or Macro & Micro-Test Viral DNA/RNA Column (HWTS-3020-50). The extraction should be processed according to the instructions for use. The extraction sample volume is 200 μL, and the recommended elution volume is 80 μL.

3、Cervical Swab/Vaginal Swab

Before sampling, use a cotton swab to gently wipe the excess secretions from the cervix, and use another cotton swab infiltrated with the cell preservation solution or the cervical exfoliated cell sampling brush to cling to the cervical mucosa and turn clockwise 3-5 rounds to obtain cervical exfoliated cells. Slowly take out the cotton swab or brush, and put it into a sample tube with 1mL of sterile normal saline. After fully rinsing, squeeze dry the cotton swab or brush against the tube wall and discard, tighten the tube cap, and mark the sample name (or number) and type on the sample tube.
